血清 LncRNA MAGI2-AS3、miR-374b-5p表达水平与前列腺癌患者 Gleason分级评分的相关性研究

摘要:
目的 探讨血清 LncRNA MAGI2-AS3、miR-374b-5p表达水平与前列腺癌患者 Gleason分级评分的相关性。方法:选取 2021年 11月— 2023年 11月于本院接受治疗的前列腺癌患者(n=110)作为前列腺癌组,另选取同期在本院体检的良性前列腺疾病患者(n=110)作为对照组。采用实时荧光定量 -聚合酶链式反应(qRT-PCR)检测血清中 LncRNA MAGI2-AS3、miR-374b-5p表达;采用 Spearman法分析血清 LncRNA MAGI2-AS3、miR-374b-5p与 Gleason分级评分的相关性;采用 Pearson法分析血清 LncRNA MAGI2-AS3与 miR-374b-5p的关系;采用多因素 Logistic回归分析影响前列腺癌发生的因素。结果:与对照组相比,前列腺癌组患者血清 LncRNA MAGI2-AS3表达水平降低(P<0.05)miR-374b-5p表达水平升高(P<0.05)。前列腺癌患者血清 LncRNA MAGI2-AS3与 miR-374b-5p表达水平呈负相关(r=-0.673,p<0.05)。多因素 Logistic回归分析显示 LncRNA MAGI2-AS3高水平是影响前列腺癌发生的保护因素(P<0.05)miR-374b-5p高水平是影响前列腺癌发生的危险因素(P<0.05)。与低危组相比,中危组、高危组血清 LncRNA MAGI2-AS3表达水,平±次降低(约0.05)miR-374b-5p表达水平±次升高(P<0.05)。前列腺癌患者血清 LncRNA MAGI2-AS3与 Gleason分级评分呈负相关(r=-0.497,p<0.05)miR-374b-5p表达水平与 Gleason分级评分呈正相关(r=0.456,p<0.05)。结论:前列腺癌患者血清 LncRNA MAGI2-AS3表达水平,显著降低, miR-374b-5p表达水平显著升高,与 Gleason分级评分有关。

Correlation between serum LncRNA MAGI2-AS3, miR-374b-5p expression levels and Gleason grading score in patients with prostate cancer

Abstract:
Objective To explore the correlation between the expression levels of serum LncRNA MAGI2-AS3 and miR-374b-5p and Gleason grading score in patients with prostate cancer. Methods Prostate cancer patients (n=110) who received treatment at our hospital from November 2021 to November 2023 were collected as the prostate cancer group, and benign prostate disease patients (n=110) who underwent physical examination were as the reference group. Real-time fluorescence quantitative polymerase chain reaction (qRT-PCR) was applied to detect the expression of LncRNA MAGI2-AS3 and miR-374b-5p in serum; Spearman method was applied to analyze the correlation between serum LncRNA MAGI2-AS3, miR-374b-5p and Gleason grading score; Pearson method was applied to analyze the relationship between serum LncRNA MAGI2 -AS3 and miR -374b -5p;multivariate Logistic regression was applied to analyze the factors influencing the occurrence of prostate cancer. Results Compared with the reference group, the serum LncRNA MAGI2-AS3 expression level in the prostate cancer group decreased (p<0.05), while the miR-374b-5p expression level increased (p<0.05). The serumLncRNA MAGI2-AS3 and miR-374b-5p expression levels in prostate cancer patients were negatively correlated(r=-0.673, <0.05). Multivariate Logistic regression analysis showed that LncRNA MAGI2-AS3 was a protective factor affecting the occurrence of prostate cancer (p<0.05),and miR -374b -5p was a risk factor affecting the occurrence of prostate cancer (p<0.05). Compared with the low -risk group, the serum LncRNA MAGI2 -AS3 expression level in the medium and high-risk groups decreased sequentially (p<0.05), and the miR-374b-5p expression level increased sequentially (p<0.05). The serum LncRNA MAGI2-AS3 in prostate cancer patients was negatively correlated with Gleason grading score (r=-0.497, p<0.05), and the expression level of miR-374b-5p was positively correlated with Gleason grading score (r=0.456, p<0.05). Conclusion The expression level of serum LncRNA MAGI2-AS3 in prostate cancer patients is greatly reduced, while the expression level of miR-374b-5p is greatly increased, which are related to Gleason grading score.
