去甲斑蝥素抑制基质金属蛋白酶-9蛋白表达对前列腺癌PC-3细胞迁移及侵袭力的影响

摘要:
目的:研究去甲斑蝥素抑制基质金属蛋白酶(MMP)-9蛋白表达对前列腺癌PC-3细胞迁移及侵袭能力的影响。方法:将去甲斑蝥素作用于前列腺癌PC-3细胞后,Western-blot和实时荧光定量PCR检测MMP-9表达水平的变化,划痕实验和侵袭实验检测PC-3细胞侵袭力的变化,透射电镜观察PC-3细胞的伪足形态变化。结果:去甲斑蝥素干预组PC-3细胞中MMP-9的mRNA表达量显著低于空白对照组(P<0.05),同时,去甲斑蝥素干预组PC-3细胞中MMP-9的蛋白表达量显著低于空白对照组(P<0.05);迁移与侵袭实验结果显示,去甲斑蝥素干预组PC-3细胞划痕愈合程度较空白对照组显著降低(P<0.05);去甲斑蝥素干预组PC-3细胞侵袭能力显著低于空白对照组(P<0.05),透射电镜观察发现去甲斑蝥素干预组PC-3细胞的伪足较空白对照组明显减少。结论:去甲斑蝥素作用于前列腺癌PC-3细胞后,PC-3细胞的侵袭能力下降,可能与MMP-9蛋白表达水平下降使PC-3细胞的侵袭伪足减少有关。

Effect of MMP-9 Expression Inhibition by Norcantharidin Interference on Migratory and Invasive Ability of PC-3 cells

Abstract:
Objective To study the effect of norcantharidin on the invasion of PC-3 cells. Methods When norcantharidin was applied to PC-3 cells, Western blot and real-time fluorescence quantitative PCR were used to detect the expression level of MMP-9.The scratch test and invasion test were used to detect the invasiveness of PC-3 cells, and transmission electron microscopy was used to observe the morphological changes of invasive pseudopods of PC-3 cells. Results The mRNA expression of MMP-9 in PC-3 cells of norcantharidin intervention group was significantly lower than that in PC-3 cells of blank control group. The expression of MMP-9 in PC-3 cells of the norcantharidin intervention group was significantly lower than that of the blank control group (P<0.05).The scratch test results showed that the scratch healing degree of PC-3 cells in the norcantharidin intervention group was significantly lower than that of the blank control group (P<0.05). The invasion ability of PC-3 cells in the norcantharidin intervention group was significantly lower than that of the blank control group (P<0.05). The transmission electron microscope observation showed thatthe invasion of pseudopodia by PC-3 cells in the norcantharidin intervention group was significantly less than that in the blank control group. Conclusion After norcantharidin acts on PC-3 cells, the invasion of PC-3 cells decreases, which may be related to the decrease of the expression level of MMP-9.
