加味启膈散抑制细胞增殖治疗 Barrett食管作用机制研究

摘要:
目的:观察加味启膈散对 Barrett食管 (BE)模型大鼠 PCNA、Ki-67 蛋白表达的影响。方法: SPF级成年健康 Wistar大鼠 72只,雌雄各半,常规适应性喂养 7天后,随机分为 6组:正常组、模型组、全方组、行气活血组、理气化痰组、化痰祛瘀组,每组 12只。采用改良后的“食管十二指肠端侧吻合加铁剂术”进行 BE造模。造模 31周后,正常组、模型组大鼠给予生理盐水灌胃,各治疗组分别给予相应药液灌胃,连续给药 14天后,处死动物留取吻合口食管组织,采用免疫组化法检测 PCNA、Ki-67蛋白表达情况。结果:与正常组比,模型组大鼠 Ki-67表达显著增强;全方组 Ki-67 表达低于模型组( P<0.05);理气化痰组和行气活血组 Ki-67表达水平高于全方组( P<0.05),化痰祛瘀组 Ki-67表达与全方组差异无统计学意义(P>0.05)。模型组 PCNA表达水平较正常组显著增强(P<0.05);全方组 PCNA表达水平显著低于模型组(P<0.05);理气化痰组和行气活血组 PCNA表达均高于全方组,差异有统计学意义( P<0.05),化痰祛瘀组与全方组 PCNA表达差异无统计学意义(P>0.05)。结论:加味启膈散抑制细胞增殖治疗 Barrett食管,其机制与抑制 PCNA、Ki-67 蛋白表达有关。

Research on Mechanism of Jiawei Qige San Inhibits Cell Proliferation in the Treatment of Barrett' s Esophagus Action

Abstract:
Objective To observe the effect of Qige San onthe PCNA andKi-67 protein expression of BE model rat. Methods Seventy-two clean adult Wistar rats, male and female, were randomly divided into 6 groups, 12 rats in each group, which were normal control group, model control group, and traditional Chinese medicine group. The whole traditional Chinese medicine group divided into the Chinese medicine Qi and activating blood group, the Chinese medicine qi stagnation group, the Chinese medicine phlegm group. Except the normal group, the other ?ve groups were modeled on BE. After successful modeling for 31 weeks, rats in the normal control group and the model control group were intragastrically administered with normal saline from the beginning of the 32nd week. Rats in each treatment group were given the corresponding liquid weight. After 14 days of treatment, the animals in each group were sacri?ced for index detection. Results For the expression of Ki-67, compared with the normal group, the expression levels of Ki-67 in the model group, the Chinese medicine Qi stagnation group, the Chinese medicine qi stagnation group and the Chinese medicine phlegm group were higher than those of the normal group. The differences were statistically signi?cant (P<0.05). Compared with the model group, the expression of Ki-67 in the whole group was lower than that of the model group, which the difference was statistically signi?cant (P<0.05). Compared with the whole group, the expression of Ki-67 in the Chinese medicine Liqi Huatan group and the Chinese medicine Xingqi Huoxue group was higher than those of the whole group and the difference was statistically significant (P < 0 . 05 ). Compared with the normal group, the expression of PCNA in the model group, the Chinese medicine qi and blood group and the Chinese medicine phlegm group were higher than that in the normal group and the difference was statistically signi?cant.The signi?cance of the study was P<0.05. The difference between the whole group and the normal group was not statistically signi?cant (P>0.05).Compared with the model group, the PCNA expression level of the whole group was lower than that of the model group. The difference was statistically signi?cant (P<0.05). Compared with the whole group, the expression of PCNA in the Chinese medicine Liqi Huatan group and the Chinese medicine Xingqi Huoxue group was higher than that in the whole group, the difference was statistically signi?cant (P<0.05).The expression of PCNA in the Chinese medicine phlegm group was higher than that in the whole group, but the difference was not statistically signi?cant (P>0.05).For the Chinese medicine qi sputum group, Chinese medicine qi and blood circulation group, there were no significant differences between the Chinese medicine and phlegm groups (P>0.05). Conclusion Jiawei QigeSan inhibits cell proliferation and treats Barrett’s esophagus. Its mechanism is related to the inhibition of PCNA and Ki-67 protein expression.
