红藤汤对小鼠溃疡性结肠炎的保护作用及机制探究

摘要:
目的:观察红藤汤对葡聚糖硫酸钠(dextran sulphate sodium, DSS) 诱导的溃疡性结肠炎小鼠的抗炎和抗氧化作用,分析红藤汤对溃疡性结肠炎的保护机制。方法:SPF 级BALB/c 小鼠30 只,随机分成3 组(n =10):对照组(C 组)、DSS诱导的溃疡性结肠炎模型组(M 组)、红藤汤治疗组(H 组)。C 组自由饮水,M 组和H 组采用自由饮用4% 的DSS 建立溃疡性结肠炎模型,H 组从造模第1 天起开始给药,连续给药10 天后,采集动脉血检测TNF-α 和IL-6 的浓度,行结肠组织病理学评分,比色法测定MDA 量及SOD 和GSH-Px 活性,Western blotting 检测ERK1/2、P-ERK1/2、JNK、P-JNK 及GADPH 蛋白的表达量。结果:与M 组比较,H 组的结肠病理学评分和血清中炎症因子(TNF-α 和IL-6)含量显著降低(P <0.05);结肠组织中MDA 的含量降低(P <0.05),而SOD 和GSH-Px 活性升高(P <0.05), P- ERK1/2 和P-JNK 的表达水平也明显下调(P <0.05)。结论:红藤汤对DSS 诱导的小鼠溃疡性结肠炎有保护作用,可以上调抗氧化基因,减少自由基的生成,重建氧化还原平衡,其作用机制可能与抑制JNK 和ERK1/2 信号通路的活化,减少炎症因子生成有关。

Abstract:
Objective To observe the anti-inflammatory and anti-oxidative effects of Hongteng Decoction mice with ulcerative colitis induced by Dextran sulphate sodium (DSS), and to clarify the protective mechanism of Hongteng Decoction on ulcerative colitis. Methods Thirty healthy and clean BALB/c mice were randomly divided into three groups (n =10): normal control group (group C), DSS-induced ulcerative colitis model group(group M), and model + Hongteng Decoction group (group H). Group C was free to drink water. Group M and Group H were freely drinking 4% DSS to establish ulcerative colitis model. Group H was administered from the first day of modeling. After 10 days of continuous administration, arterial blood was collected for TNF-a and the concentration of IL-6, colon histopathological score, colorimetric determination of MDA amount and SOD and GSH-PX activity, Western blot detection of ERK1/2, P-ERK1/2, JNK, P-JNK and GADPH protein Expression. Results Compared with group M, colonic pathology scores and serum inflammatory factors(TNF-α and IL-6) levels were significantly lower in group H (P <0.05); MDA level in colon tissues was decreased (P <0.05), while SOD and the activity of GSH-PX increased (P <0.05), and the expression levels of P-ERK1/2 and P-JNK were also significantly down-regulated (P <0.05). Conclusion Hongteng Decoction has protective effect on DSS-induced ulcerative colitis in mice. It can up-regulate antioxidant genes, reduce free radical production, re-establish the redox balance. In addition,Hongteng Decoction also inhibits the activation of JNK and ERK1/2 signaling pathways, reduces the production of inflammatory factors, and eventually inhibits
