目的 观察异钩藤碱(ISO)对非小细胞肺癌(NSCLC)细胞的增殖和迁移的影响,并探讨其作用机制。方法:选择人NSCLC细胞系A549,随机分为对照组和ISO组,分别用0和2 mg/L浓度的ISO处理A549细胞24 h,通过CCK-8和克隆形成实验评估细胞生长及增殖状态,通过划痕实验评估细胞运动和迁移能力,通过免疫印迹和铁检测试剂盒评估细胞铁死亡,并通过免疫印迹实验检测Wnt/β-catenin信号通路的激活状态。结果:与对照组比较,ISO组A549细胞的增殖和迁移能力降低,铁死亡增加,WNT3α表达以及β-catenin、GSK-3β的磷酸化水平降低。结论:ISO具有抑制NSCLC细胞增殖和迁移并促进铁死亡的作用,其机制可能与抑制Wnt/β-catenin信号通路激活有关。 |

Abstract: |
Objective To investigate the effects of isorhynchophylline on the growth, motility, and ferroptosis of non-small cell lung cancer (NSCLC) cells, as well as its influence on the Wnt/β-catenin signaling pathway. Methods The human NSCLC cell line A549 was selected for grouped experiments. A549 cells were treated with 0 and 2 mg/L concentrations of isorhynchophylline for 24 hours. Cell growth was evaluated using CCK-8 and clonogenic assays, while cell motility was assessed through wound healing assays. Ferroptosis was measured using immunoblotting and iron detection kits, and the activation of the Wnt/β-catenin signaling pathway was evaluated by Western blotting. Results Isorhynchophylline significantly inhibited the growth and motility of human NSCLC cells. Additionally, isorhynchophylline promoted ferroptosis in NSCLC cells. The antitumor effects of isorhynchophylline were associated with the inhibition of the Wnt/β-catenin signaling pathway, which is crucial for cell survival and growth. Conclusion: Isorhynchophylline exerts antitumor effects on NSCLC cells by inhibiting cell growth and motility, promoting ferroptosis, and suppressing the Wnt/β-catenin signaling pathway. |
