目的 比较五达颗粒和爱维莫潘对术后肠麻痹( POI)模型小鼠胃肠移行性复合运动( MMC)的作用。方法:在 15只成年 C56BL/6小鼠的胃窦、十二指肠浆膜表面植入微型应力传感器,随机分为对照组、 POI-五达颗粒和 POI-爱维莫潘组,每组各 5只,比较不同剂量五达颗粒(0.75、1.5及 3 g/kg)和爱维莫潘(0.25、0.5和 1 mg/kg)对 POI小鼠术后胃肠 MMC的影响。结果:对照组小鼠胃窦、十二指肠出现典型的 MMC,POI小鼠胃窦、十二指肠 MMC周期紊乱,出现不规则收缩波,与对照组比较, POI小鼠收缩振幅、频率和振幅指数均下降,差异有统计学意义(p<0.001)。±次给予 0.75、1.5及 3 g/kg五达颗粒后, POI小鼠胃肠运动显著增强,可诱发高频率高振幅的MMC芋相样收缩,收缩频率、振幅和振幅指数与给药前基础值相比,均有明显增加,差异有统计学意义(p<0.001)。±次给予 0.25、0.5和 1 mg/kg爱维莫潘并未明显增强胃窦、十二指肠收缩能力,收缩频率、振幅和振幅指数与给药前相比无显著差异。结论:肠道操作导致小鼠胃肠 MMC障碍,五达颗粒通过诱导MMC芋相样收缩而改善术后胃肠运动功能,爱维莫潘不是通过直接增强术后胃肠运动来治疗 POI。 |

Comparison of the effects of Wuda granules and alvimopan on gastrointestinal migrating motor complex in postoperative ileus model mice |

Objective To observe the effect of Wuda granules and alvimopan on gastrointestinal migrating motor complex (MMC) movement in postoperative ileus model mice. Methods 15 adult C56BL/6 mice were implanted with miniature stress sensors on the surface of the gastric antrum and duodenal serosa, and randomly divided into a control group, POI-Wuda granules group, and POI-Avimopan group, with 5 mice in each group, to compare the effects of Wuda granules and Avimopan on postoperative gastrointestinal MMC motility in POI mice. Results The control group of mice showed typical MMC in the gastric antrum and duodenum. After intestinal manipulation, the MMC cycle in the gastric antrum and duodenum of mice was disrupted, and irregular contractile waves appeared. Compared with the control group, the amplitude, frequency, and amplitude index of contractions decreased (p<0.001). Sequential administration of Wuda granules (0.75, 1.5 and 3 g/kg) induced high-frequency and high amplitude MMC III phase like contractions, with significant increases in contraction frequency, amplitude, and amplitude index compared to the baseline values before administration (p<0.001).Sequential administration of alvimopan (0.25, 0.5 and1 mg/kg) did not significantly enhance gastric antral and duodenal contractions, with no significant differences in contraction frequency, amplitude, and amplitude index compared to before administration. Conclusion Intestinal manipulation leads to impaired gastrointestinal MMC motility in mice. Wuda granules improve postoperative gastrointestinal motility by inducing MMC phase 芋contraction, while the effect of alvimopan in treating POI is not directly enhanced by postoperative gastrointestinal motility. |
