Effect of Zhushi Tuoju recipe on inflammatory factors and bacterial infection of diabetes ulcer wound in rats

Abstract:
Objective To observe the effect of Zhushi Tuoju recipe on diabetes ulcer in rats, and compare the changes of inflammatory factors and bacterial infection levels of the wound before and after treatment. Methods A total of eighty 10 weeks old Wistar rats were randomly assigned to two groups: a blank control group(n=20) and a model preparation group (n=60). The model preparation group then underwent induction of diabetic ulcer. Subsequently, the successfully modeled rats were randomly divided into three groups (n=20 per group): model group, control group, and treatment group. In the treatment group, local debridement, external application of epidermal growth factor gel, external application of Zhushi Tuoju recipe oil, and dressing with sterile gauze; In the control group, local debridement, external application of epidermal growth factor gel, external application of sterile gauze, and dressing with sterile gauze; Local debridement of the model group, application of sterile oil gauze, and bandaging with sterile gauze; the blank group will not be treated. On the 16th day of treatment, measure and compare the ulcer area, wound colony concentration, and inflammatory factor levels of each group. Results Compared with the control group after treatment, the treatment group showed a reduction in ulcer area, a decrease in IL-6 levels and TNF-α levels, an increase in IL-10 levels and IL-12 levels, and a decrease in wound bacterial count levels. The improvement degrees of treatment group were better than that of the control group, and there were a statistical difference (P <0.05). Conclusion It is preliminarily proved that the external application of Zhushi Tuoju recipe oil can improve the wound area of rats before and after treatment, reduce the level of proinflammatory factors, increase the level of anti-inflammatory factors, and reduce the bacterial count. Its therapeutic effect may be related to the effective ingredients in the formula, the regulation of inflammatory factors, and the direct killing of pathogenic microorganisms.
