基于RAS/RAF/ERK信号通路探讨益气生肌合剂对压力性损伤大鼠创面微小血管生成的作用机制

摘要:
目的:观察益气生肌合剂通过调控RAS/RAF/ERK信号通路对压力性损伤大鼠模型创面修复及微小血管生成的影响。方法:32只大鼠分为模型对照组、三乙醇胺乳膏组(阳性药物组)、益气生肌合剂组(益气生肌组)、三乙醇胺乳膏+益气生肌组(联合用药组),每组8只。建立压力性损伤大鼠模型并予不同方式干预。记录各组大鼠创面愈合及病理损伤情况,检测各组大鼠创面血流灌注量水平及微血管密度(MVD),Western blotting法及RT-PCR法检测创面组织中Ras、Raf1、ERK1/2及VEGF的蛋白及mRNA表达情况。结果:与模型对照组比较,阳性药物组、益气生肌组、联合用药组大鼠压力性损伤创面均呈明显愈合趋势,其中联合用药组大鼠的创面愈合率最高,差异有统计学意义(P<0.05);HE染色结果亦证实联合用药组的病理缓解程度最佳;益气生肌合剂及三乙醇胺乳膏均能够有效促进大鼠创面的血流灌注量及微小血管生成,其中联合用药组效果最佳,差异有统计学意义(P<0.05);Western blotting及RT-PCR结果显示,阳性药物组、益气生肌组、联合用药组均可显著提升大鼠创面受压组织的Ras、Raf-1、ERK1/2及VEGF的蛋白表达及mRNA表达,且联合用药组效果最优,差异有统计学意义(P<0.05)。结论:益气生肌合剂能有效促进压力性损伤大鼠模型创面修复,同时与三乙醇胺乳膏联合使用起到协同增效作用,其作用机制可能与益气生肌合剂能够激活RAS/RAF/ERK信号通路并促进该信号通路相关蛋白表达相关。

Based on RAS/RAF/ERK Signaling Pathway to Explore the Mechanism of YiQi ShengJi Mixture on the Formation of Microangiogenesis in the Wound Surface of a Pressure-Injured Rat Model

Abstract:
Objective To observe the effect of Yiqi Shengji Mixture on wound repair and microangiogenesis in a rat model of pressure injury by regulating the RAS/RAF/ERK signaling pathway. Methods A rat model of stress injury and intervene was established in different ways. The wound healing and pathological damage of rats were recorded in each group, then the blood perfusion level and microvessel density (MVD) of the wound were detected in each group. Western blotting method and real-time quantitative PCR detection method was used to detect Ras and Raf-1 in the wound tissue, ERK1/2, VEGF and other proteins and mRNA expression. Results Compared with the model control group, the pressure-injured wounds of the rats in the positive drug group, the Yiqishengji group and the combined medication group all showed a significant healing trend. Among them, the combined medication group had the highest wound healing rate (P < 0.05). HE staining results also confirmed that the pathological remission of the combined medication group was the best (P < 0.05). Both Yiqi Shengji mixture and triethanolamine cream could effectively promote the blood perfusion and microangiogenesis in the wounds of rats. Among them, the combined drug group had the best effect (P < 0.05).
