B7-H1 与B7-H4 在膀胱癌大鼠及其癌旁组织中的差异表达

摘要:
目的:研究膀胱原位癌组织及癌旁组织中共刺激分子 B7-H1与 B7-H4的表达情况,并分析二者的相关性。方法:采用 N-亚硝基 -N-甲基脲(MNU)溶液膀胱灌注法制备膀胱癌大鼠模型(30只),处死大鼠,获取癌组织及癌旁组织,进行病理检查;采用生理盐水溶液膀胱灌注法作为对照组( 12只),处死大鼠,获取膀胱组织,进行病理学检查;采用 RT-PCR技术分别检测对照组大鼠膀胱组织( A组)、膀胱癌模型组癌组织( B组)及模型组癌旁组织( C组)中 B7-H1与 B7-H4的 mRNA的表达水平,并分析二者之间的相关性。结果: 30只膀胱癌模型组大鼠, 28只模型制备成功,造模成功率 93.3%。A组、C组 B7-H1、B7-H4 mRNA表达量均明显低于 B组(0.71±0.38、0.79±0.21 vs 1.44±0.67;0.84±0.22、

Establishment of Bladder Cancer Model in Rats and Differential Expression of B7-H1 and B7-H4 in Cancer Tissue

Abstract:
Objective To investigate the expression of B7-H1 and B7-H4 in tumor and paracancerous tissues of bladder cancer rats and analyze their correlation. Methods 30 rat models of bladder cancer were prepared by bladder perfusion with MNU solution, and 28 rats were successfully established. The rats were killed to obtain tumor tissue and precancerous tissue for pathological examination. The contents of B7-H1 and B7-H4 in bladder tissue (group A, n=12), tumor tissue (group B, n=28) and precancerous tissue (group C, n=28) of 12 non bladder cancer rats were detected by RT-PCR, and the correlation between them were analyzed. Results In 30 rats, 28 models were successfully established, with a success rate of 93.3%. The RNA content of B7-H1 and B7-H4 in group A and group C were signi.cantly lower than those in group B (0.71±0.38, 0.79±0.21 vs 1.44±0.67; 0.84±0.22, 0.92±0.31 vs 1.87±0.37), which the difference was statistically signi.cant. The content of both in group C was slightly higher than that in group A, but there was no statistical signi.cance (P > 0.05). There was a positive correlation between B7-H1 and B7-H4 RNA content (r=0.908, P < 0.05). Conclusion MNU solution intravesical instillation can successfully prepare bladder cancer rat model. B7-H1 and B7-H4 are both highly expressed in bladder cancer tissue. Both can be used as new tumor markers and potential therapeutic targets for bladder cancer, which providing new ideas for clinical diagnosis and treatment.
