丹酚酸B 对糖尿病小鼠创面愈合及局部碱性成纤维细胞生长因子、转化生长因子-β1 的影响

摘要:
目的:探讨丹参主要成分丹酚酸B(Sal B)对糖尿病小鼠慢性伤口创面修复的影响及其分子机制。方法:将30只db/db糖尿病小鼠随机分为模型组、Sal B组(1 mg/mL)、碱性成纤维细胞生长因子(bFGF)组(3.6 μg/mL),每组10只;另取db/m小鼠8只为空白组。各组小鼠背部全厚皮切除术制备伤口模型,空白组和模型组外用含生理盐水的明胶海绵;bFGF组外用含bFGF的明胶海绵,Sal B组外用含Sal B的明胶海绵。外用药以一次性无菌敷料贴包扎,隔日换药1次,并拍摄创面照片计算愈合率,给药周期为14 d。实验结束后取创面皮肤,采用苏木素-伊红(HE)染色,观察病理学改变,采用实时定量PCR法和Western blotting检测创面局部生长因子bFGF、转化生长因子β1(TGF-β1)基因和蛋白的表达。结果:造模第3、7、14天,模型组创面愈合率分别为(0.26±0.11)%、(0.43±0.12)%、(0.66±0.12)%,低于空白组[(0.63±0.09)%、(0.81±0.97)%、(0.98±0.02)%],bFGF组创面愈合率[(0.37±0.12)%、(0.59±0.07)%、(0.79±0.04)%]与Sal B组创面愈合率[(0.40±0.05)%、(0.58±0.05)%、(0.75±0.06)%]高于模型组(均P<0.05)。模型组bFGF mRNA、TGF-β1mRNA的表达水平分别为(0.19±0.05)、(0.32±0.08),低于空白组[(0.95±0.16)、(1.35±0.31)],差异有统计学意义(P<0.05),bFGF组TGF-β1 mRNA的表达水平(1.31±0.33)高于模型组(P<0.01),Sal B组bFGF、TGF-β1 mRNA的表达[(0.71±0.25)、(1.42±0.48)]均高于模型组(P<0.05)。模型组bFGF、TGF-β1蛋白的表达[(0.38±0.13)、(0.26±0.12)]低于空白组[(0.58±0.05)、(0.55±0.16)](P<0.05),bFGF组bFGF、TGF-β1蛋白表达[(0.64±0.12)、(0.56±0.16)]均高于模型组,Sal B组bFGF、TGF-β1蛋白表达[(0.63±0.10)、(0.68±0.07)]高于模型组(P<0.05)。结论:Sal B可以促进糖尿病小鼠创面愈合,其机制可能是升高创面生长因子的水平,促进肉芽组织生成。

Effects of Salvianolic Acid B on Wound Healing and the Expression of Local bFGF and TGF-β1 in Diabetic Mice

Abstract:
Objective To explore the effect of salvianolic acid B (Sal B), the main component of Salvia miltiorrhiza, on wound healing of chronic wounds in diabetic mice and its molecular mechanism. Methods The experiment was divided into 4 groups, 30 db/db diabetic mice were randomly divided into model group, Sal B group and basic fibroblast growth factor (bFGF) group, 10 mice in each group. The control group consisted of 8 db/m mice. Wounds were prepared by full thickness skin resection on the back of mice in each group. The control group and the model group were externally used gelatin sponge containing normal saline. The bFGF group was externally used gelatin sponge containing bFGF and the Sal B group was externally used gelatin sponge containing Sal B. The external medication were wrapped with a disposable sterile dressing, the dressing were changed every other day and the wounds were taken to calculate the healing rate. The experimental period was 14 days. After the experiment, the wound skin was taken and stained with hematoxylineosin (HE) to observe the pathological changes. The expression of bFGF and transforming growth factor β1(TGF β1) genes and proteins were detected by real-time quantitative PCR and Western blotting protein quantitative methods respectively. Results On the 3rd, 7th, and 14th days after wound formation, the wound healing rate of the model group [(0.26±0.11)%, (0.43±0.12)%, (0.66±0.12)%] was lower than those of the control group [(0.63±0.09)%, (0.81±0.97)%, (0.98±0.02)%] (P<0.05). The wound healing rate of the bFGF group [(0.37±0.12)%, (0.59±0.07)%, (0.79±0.04)%] and Sal B group [(0.40±0.05)%, (0.58±0.05)%, (0.75±0.06)%] were higher than those of the model group (P<0.05). Compared with the control group [(0.95±0.16), (1.35±0.31)], the expression levels of bFGF mRNA and TGF-β1 mRNA in the model group [(0.19±0.05), (0.32±0.08)] were decreased (P<0.05). Compared with the model group, the expression of TGF-β1 mRNA in the bFGF group (1.31±0.33) was increased (P<0.01), and the expression of bFGF and TGF-β1 mRNA in the Sal B group [(0.71±0.25), (1.42±0.48)] were both increased (P<0.05). In addition, compared with the model group [(0.38±0.13), (0.26±0.12)], the expression of bFGF and TGF-β1 protein in the control group [(0.58±0.05), (0.55±0.16)], bFGF group [(0.64±0.12), (0.56±0.16)] and Sal B group [(0.63±0.10), (0.68±0.07)] were all increased (P<0.05). Conclusion Sal B can promote wound healing in diabetic mice. The mechanism may be to increase the level of wound growth factors and promote granulation tissue formation.
