肝细胞癌中组蛋白甲基化酶SET结构域分支型1的表达及其临床意义

摘要:
目的:分析肝细胞癌中组蛋白甲基化酶SET结构域分支型1(SETDB1)的表达及其临床意义。方法:选取90例肝细胞癌患者,分别采集其肝细胞癌组织和癌旁组织,采取qRT-PCR法对其中SETDB1的水平进行检测,并进行组间对比分析。结果:肝细胞癌组织中SETDB1阳性率明显高于癌旁组织(P<0.05);肝细胞癌组织中SETDB1相对表达量明显高于癌旁组织(P<0.05),肿瘤直径为6 cm及以上者的癌组织SETDB1水平明显高于肿瘤直径不足6 cm者(P<0.05);MVI分型中,M1+M2型的癌组织SETDB1水平明显高于M0型(P<0.05);pTNM分期中,Ⅲ+Ⅳ期患者癌组织SETDB1水平明显高于I +II期(P<0.05)。结论:肝细胞癌中组蛋白甲基化酶SETDB1的水平明显升高,且肿瘤直径较大、M1+M2型、pTNM分期较高患者的肝细胞癌内SETDB1的水平明显升高。

Expression of Histone Methylase SETDB1 in Hepatocellula Carcinoma and its Clinical Significance

Abstract:
Objective To analyze the expression of histone methylase SETDB1 in hepatocellular carcinoma and its clinical significance. Methods 90 patients with hepatocellular carcinoma were selected and from which 90 hepatocellular carcinoma tissues and 90 paracancerous tissues were collected. The qRT-PCR method was adopted to detect the level of SETDB1 among them and to expand the comparison between groups. Results The positive rate of SETDB1 in cancer tissues was significantly higher than that in adjacent tissues (P<0.05). The relative expression of SETDB1 in hepatocellular carcinoma tissues was significantly higher than that in adjacent tissues (P<0.05). The tumor diameter was 6 cm and above. The level of SETDB1 in cancer cells was significantly higher than that of tumors less than 6 cm in diameter (P<0.05). In MVI classification, the level of SETDB1 in cancer cells of M1+M2 stage was significantly higher than that of M0 stage (P<0.05). In pTNM staging, the level of SETDB1 in cancer cells of stage Ⅲ+Ⅳ was significantly higher than that in stage I+II (P<0.05). Conclusion The level of histone methylase SETDB1 in hepatocellular carcinoma was significantly increased, and the tumor diameter was larger. M1+M2 stage and pTNM stage were relatively large. In patients with high levels, the level of histone methylase SETDB1 in hepatocellular carcinoma significantly increased.
